Real-time protection is ok.Do you use any special settings?

Additional On-demand scanners:
Emsisoft Emergency Kit (Free) - link
With the Emsisoft Emergency Kit Scanner you have got the powerful Emsisoft Scanner including graphical user interface. Search the infected PC for Viruses, Trojans, Spyware, Adware, Worms, Dialers, Keyloggers and other malign programs.
Run the Emsisoft Emergency Kit Scanner with a double click on a2emergencykit.exe. Found Malware can be moved to quarantine or finally deleted.

Additional browser protection:
WOT (Free) - link
To help you avoid malicious sites you can use Web of Trust (WOT) a website rating browser plugin. After you add it to your browser make sure you only visit websites rated "Green" by WOT

VTzilla (Free) - link
VTzilla is a Mozilla Firefox browser plugin that simplifies the process of scanning Internet resources with VirusTotal. It allows you to download files directly with VirusTotal's web application prior to storing them in your PC. Moreover, it will not only scan files, but also URLs.
The scanning options are embedded in Firefox's context menu and download dialog, making the analysis process as easy as clicking a single button.

CIS is a comprehensive security suite so you're pretty much set. Just don't give up on basic principles like staying away from unknown executables or updating OS & software, thinking that CIS will keep you safe no matter what. Full image backups of your hdd/system partition are also a must. I use Paragon Backup & Recovery 2010 Free Advanced.

An external hard-drive with eSATA or USB3 connection is the best option. You'll also need to create a bootable recovery USB/CD containing the backup program, in case you can't start Windows (in Paragon go to Tools -> Recovery media builder).

Backing up on a second partition or a second internal HDD is better than nothing but it doesn't cover situations like HDD failure or malware that affects the MBR, possibly braking the partition table. But i suppose you can backup just the MBR on a usb stick/CD.

If space on your external drive is an issue you can create multiple partitions, the first one for your operating system and programs and another one for large data files such as movies, music, pictures or even games (since games today use up allot of GB). You will need a full image of your first partition (with the OS and apps). As for the data files you can backup just the important ones.

It is recommended to use online storage services like Dropbox, iDrive, Carbonite, etc. for important data files. Some offer free space and If you convince your friends to join these services they'll usually give you extra space for free. You can get up to 8GB for free on Dropbox. This can be sufficient for important files.
Here is a nice article about backup: link. It talks about best practices when you backup important files.
