comodo help says default enabled so ı think checked > enabled / unchecked > disabled

Keep it enabled, you won't like a malware injecting code in a process lol

If you have a conflict with a certain software, you can add that process to the exceptions list.
Shellcode injection protection is mainly for Comodo self-protection.

As @shmu26 said. only if there is a conflict with a given program or you want to exclude it, you have to keep it activated.
