My PC's main line of protection is Deep Freeze
http://www.faronics.com/products/deep-freeze/standard/

Protected against
-MBR Modifiers
-Cryptolockers
-Ransomeware
-RATS
-Ring 0,1,2,3 Root Kits
-Viruses aimed to delete important system files, mess with drive formatting

I have a variety of backup options incase the actual protection line fails.

Any Important Documents I make are transferred to a separate portable hard drive and deleted from the main PC.

If i ever leave my PC unattended (my main one)

I leave Faronics Anti Executable Open http://www.faronics.com/products/anti-executable/
-Basically won't allow anything to be open that is an executable file lel

Well that's considered another good thing running at a fully virtualized desktop, I guess its already a good configuration. ;)

Perhaps adding adblocker for Chrome like Adblock Plus or Adguard is fine for uninterrupted browsing experience. :)

You should consider using these add-ons for google chrome:
  • Adblock Plus
  • WOT (Web of Trust)
  • HTTPs Everywhere
  • ScriptSafe

Ahh, forgot to mention chrome addons

-LastPass (Password Manager) I have to login to it to use it, every restart so viruses don't take the pass. Well, you need the google authenticator code and my cell phone to actually use it on a different HWID
-Rumla (Capcha AutoSolver)
-Adblock
-HTTPS Everywhere

(Also I do have service pack 1 on my pc, and am on latest windows updates)
