Does Kaspersky's HIPS protect against like things trying to destroy the system?
If you are talking about disk wipers, Kaspersky can protect you from these by blocking untrusted apps and scripts. If you keep all your apps updated and to the minimum (do not install anything not needed), the possibility of something managing to sneak in and wipe your disk is very low. To find out how to configure Kaspersky to block untrusted apps and scripts, use one of the guides that @harlan4096 linked.

Have a default deny setup. But does Kaspersky protect core applications from malware?

Yes it does, you should take a look at it and atleast configurate the "Rights Tab" for High and Low Restricted. Deactivating Code Injection and denying Acess to Shutdown Windows and Denying Access do KLDriver is a good start. But i would also configurate the File System Part in Protected Applications and deny all acess to cmd and powershell for superior protection.You can do hella lot with Kaspersky's HIPS System, and its actually really easy, surely it takes a bit of time but everyone with basic knowledge can lockdown the system if wished.
Its only important to NOT change any setting for the trust group. Because in theres everything, From Windows Apps, Processes to Kasperskys stuff itself, it will break stuff.
